Human, who is at the club on loan, missed the two Parker Pen Challenge Cup matches against L'Aquila because he is not Europeanregistered, but comes straight back into the starting line-up against the Falcons.
Wylie's inclusion is one of ten changes to the side that defeated L'Aquila 50-0 last weekend.
David Flatman, Jonathan Humphreys (capt), Duncan Bell, Steve Borthwick, Rob Fidler, Andy Beattie, Michael Lipman and Zak Feaunati all return in the pack, whilst Andrew Higgins has recovered from a hamstring strain to take the injured Iain Balshaw's place on the wing.
Both Mike Catt (hamstring) and Iain Balshaw (calf) miss out through injury.
"Newcastle are a very good side indeed," said Head Coach John Connolly. "They have plenty of experience in the pack where Mark Andrews and Garath Archer are very tough players. They also have plenty of pace out wide."
"It is great to have Wylie back as he has been extremely effective for us this season. Michael Lipman is back from suspension, but the pack is very much the same eight that has taken us through the first half of the season."
