Simpson-Daniel, who missed out on a World Cup ticket to Australia, returned to training earlier this week after injury – and is now fit to take his place on the left-wing.
Gloucester coach Nigel Melville said: "It's great to have an international player back, and it will certainly offer us options for Saturday."
New signing Adrian Olver starts his first game for the club after Andy Deacon was ruled out with a calf injury picked up during the defeat to London Irish last Sunday.
The prop signed for Gloucester this week on a short-term contract from Worcester and has Zurich Premiership experience with both NEC Harlequins and Saracens.
Hooker Chris Fortey is named on the bench having returned from suspension.
Looking ahead to Saturday's game, captain Jake Boer said: "Saracens are a very different side to last season. They've had a good start and they'll be happy with the results so far.
"They've got some quality players and of course the big signing for them in the summer was Taine Randell.
"Taine is a world-class player who is respected throughout the world and I'm looking forward to coming up against him on Saturday."
